Chile is considered to be a sub-center of origin for the cultivated potato, with native and introduced genetic material coexisting in the country. Thus, the different varieties present in Chiloe Island are characterized by a rich diversity of forms, sizes, colours and phenological characteristics. In the present work, the level of polymorphism and the genetic relationship were studied by means of molecular markers using the amplified fragment length polymorphism (AFLP) technique and twenty-seven morphological characters. Twenty varieties of potatoes from the Chiloe Island were analyzed. The commercial variety Desirée and one specie from the Etuberosa series, Solanum fernandezianum, collected in the Juan Fernandez Island were included as controls. A... |

For studying genetic diversity in natural populations of Terminalia, a medicinal plant, our attempts to isolate high quality DNA using several previously reported protocols and even modifications were unsuccessful. We therefore combined CTAB based isolation, and column based purification step, to isolate DNA from Terminalia arjuna. The DNA isolated using this standardized protocol was high in quality and suitable for restriction digestion and generation of random amplification of polymorphic DNA (RAPD) and amplified fragment length polymorphism (AFLP). |

Eucalyptus globulus is the second most important forest species in Chile, after Pinus radiata. The main advantages of E. globulus are its fast growth (25 m³/ha/year) and its excellent wood quality for kraft pulp production. On the negative side, its low freezing tolerance has been an obstacle for the expansion of plantations, specifically on the foothills of the Andes. The difference in the freezing resistance between clones of E. globulus has a genetic base and, therefore, it could be detected through DNA molecular markers. Fifteen clones of E. globulus, eight classified as freezing resistant and seven as freezing sensitive were analyzed using the technique of RAPD, in order to obtain molecular markers that could differentiate between freezing sensitive... |

Background Asian soybean rust (SBR) caused by Phakopsora pachyrhizi Syd. & Syd., is one of the main diseases affecting soybean and has been reported as one of the most economically important fungal pathogens worldwide. Knowledge of the genetic diversity of this fungus should be considered when developing resistance breeding strategies. We aimed to analyze the genetic diversity of P. pachyrhizi combining simple sampling with a powerful and reproducible molecular technique. Results We employed Amplified Fragment Length Polymorphism (AFLP) technique for the amplification of P. pachyrhizi DNA extracted from naturally SBR-infected plants from 23 production fields. From a total of 1919 markers obtained, 77% were polymorphic. The high percentage of... |

Background: An adaptive mechanism in plant roots is initiated in the event of nitrogen and potassium deficiency, and it facilitates the active uptake of these elements in order to ensure plant growth and survival in stress conditions. Signaling and transduction of signals in response to changing nitrogen and potassium concentrations is a complex process, affected by interactions between various gene expression products, and often subjected to modifications. Results: In order to identify genotypic differences between phenotypes of two populations of recombinant inbred rye lines (153/79-1 x Ot1-3 and Ot0-6 x Ot1-3) in response to nutrition stress caused by nitrogen and potassium deficiency at the seedling stage, bulk segregant analysis was utilized.... |

Background: Calibrachoa Cerv. (ex La Llave & Lexarza) is a genus of the Solanaceae family (La Llave and Lexarza, 1825). This genus has a high ornamental and economic value due to its intrinsic variability and multiplicity of flower colours. In Argentina there are eight native species, and one of them is Calibrachoa caesia. The genetic diversity among 35 accessions of C. caesia, from five departments in the province of Misiones, was analyzed using ISSR markers. Results: Thirteen ISSR primers yielded a reproducible banding pattern, with 701 amplified loci and 98% of polymorphism. The ISSR primers 5CT, 5CA, 5GA, 5GACA, 3CAC, 3TG and 3TC generated 100% polymorphic patterns. The Rp values ranged from 23.20 to 10.29 for 5GACA and 3AG primers,... |

Somatic embryogenesis, which is still the method of choice for tissue culture, regeneration and transformation of maize, is largely considered highly genotype-dependent. The Hi II, a highly embryogenic genotype, has been extensively used in transformation protocols. However, this is not an inbred line; instead, it has a proportion of the undesirable A-188 background, and the progeny segregates for phenotypic characteristics and shows poor agronomic performance. In an effort to identify genotypes that combine a high somatic embryogenic response and good agronomic performance, we evaluated 48 advanced inbred lines developed at INTA. Callus development and somatic embryogenesis capacity were measured in 200 immature embryos per line. Embryogenic capacity [EC... |

Amylose and amylopectin are the two polysaccharides that constitute starch in bread wheat and the enzyme GBSSI (Granule-bound starch synthase I), also known as waxy protein, is responsible for amylose synthesis in storage tissues. Decrease of the amylose content in starch has been associated with the lack of waxy protein(s). In this work, different sets of PCR markers were used to characterize the genetic variability of waxy loci from 103 Argentinean bread wheat cultivars. For the Wx-A1 locus, Wx-A1a and a novel molecular allele designed Wx-A1g were detected. Wx-B1 locus showed three alleles (Wx-B1a, Wx-B1b, Wx-B1e), and Wx-D1 locus showed only the Wx-D1a allele. Novel single-locus allele specific markers for Wx-A1b, Wx-B1b and Wx-D1b null alleles were... |

Background At present, species known as camote de cerro (Dioscorea spp.) are found only in the wilderness in Mexico, but their populations are extremely depleted because they are indiscriminately collected, it is urgent to evaluate the conservation status of these plants in order to design conservation genetics programs. In this study, genetic diversity parameters along with cluster analysis based on Jaccard's coefficient were estimated with the objective to assess the efficiency of Random Amplified Polymorphic DNA (RAPD), Inter Simple Sequence Repeat (ISSR), Amplified Fragment Length Polymorphism (AFLP) and Inverse Sequence Tagged Repeat (ISTR) molecular DNA markers in the Dioscorea genus. Results The polymorphic information contents were quite similar... |

Randomly amplified polymorphic DNA (RAPD) markers were used to estimate genetic diversity among 18 cultivars of niger from India. Total genomic DNA was extracted and subjected to RAPD analysis using 80 arbitrary 10-mer primers; 17 primers were selected, which yielded a total of 124 bands, 41.20% of them polymorphic. None of the primers produced unique banding pattern for each cultivar. RAPD data were used to calculate a Squared-Euclidean Distance matrix which revealed a minimum genetic distance between cultivars JNC-6 and N-48 and a maximum distance between IGP-76 and JN-30. Based on the distance matrix, a cluster analysis was done using a minimum variance algorithm. The dendrogram generated, based on Ward’s method, grouped 18 niger cultivars... |

In Peach (Prunus persica) several physiological changes, such as woolliness, triggered by chilling injury are involved in major production losses due to cold storage of the fruits during shipping. Additionally, the low level of polymorphisms among peach varieties is an important limitation in the search for new molecular markers that could be associated with economically important traits. Therefore, a functional approach was employed to associate candidate genes with an informative marker in peach. The data was obtained from the results of an in silico analysis of four different cold peach treatments. Thirty two candidate genes were selected that were aligned against Arabidopsis thaliana genomic sequences to design intron-flanking EST-PCR markers. These... |
